چکیده

Abstract Polyhydroxyalkanoate or PHA is produced through microbial fermentation of sugar and lipid where in nature it stored as energy carbon source for microorganisms. Most the feedstock used production are derived from biomass they remain cheapest to date. In this study, nipa palm sap was proposed an alternative since underutilized widely distributed along river estuaries Malaysia. This contains high content essential nutrients which required production. The aim work explore potential a investigate effect added non-added nutrient media on during batch using Cupriavidus necator DSM545. Extraction chloroform showed that 2 g/L recovered when with while only 2.16 PHA. All samples presence carbonyl band (C=O) extracted granules by FTIR peak obtained UV Spectrophotometer confirmed These findings proved novel

Mutations derived from the thermophilic polyhydroxyalkanoate synthase PhaC enhance the thermostability and activity of PhaC from Cupriavidus necator H16.

The thermophile Cupriavidus sp. strain S-6 accumulated polyhydroxybutyrate (PHB) from glucose at 50°C. A 9.0-kbp EcoRI fragment cloned from the genomic DNA of Cupriavidus sp. S-6 enabled Escherichia coli XL1-Blue to synthesize PHB at 45°C. Nucleotide sequence analysis showed a pha locus in the clone. The thermophilic polyhydroxyalkanoate (PHA) synthase (PhaC(Csp)) shared 81% identity with mesop...

Production of PHB from Chicory Roots – Comparison of Three Cupriavidus necator Strains

Renewable resources constitute a ‘limited infinity’, meaning that a limited amount is being produced, but for an infinite time.1 One of the most promising approaches to reduce the resource intensity is to use the existing biomass more efficiently: the biorefinery concept. It aims to use the complete biomass and all by-products emitted by the processes. Particularly the by-products of establishe...

Effect of feeding regimens on polyhydroxybutyrate production from food wastes by Cupriavidus necator.

We investigated the effects of different feeding regimens (1-pulse, stepwise, and continuous) of fermented food-waste liquid on polyhydroxybutyrate (PHB) production. The fermentation liquid was filtered with a membrane filter (pore size, 0.45 μm) to remove anaerobic microorganisms and solids and used as a carbon source for Cupriavidus necator. One-pulse feeding yielded the highest cell concentr...

Factors Affecting Poly(3-hydroxybutyrate) Production from Oil Palm Frond Juice by Cupriavidus necator (CCUG52238T)

Factors influencing poly(3-hydroxybutyrate) P(3HB) production by Cupriavidus necator CCUG52238(T) utilizing oil palm frond (OPF) juice were clarified in this study. Effects of initial medium pH, agitation speed, and ammonium sulfate (NH(4))(2)SO(4) concentration on the production of P(3HB) were investigated in shake flasks experiments using OPF juice as the sole carbon source. The highest P(3HB...

Poly(hydroxyalkanoate) Production by Cupriavidus necator from Fatty Waste Can Be Enhanced by phaZ1 Inactivation

Although PHAs are regarded as an effective substitute for conventional plastics for a number of medical and agricultural applications1 and food packaging2, their full-scale manufacturing is hampered by high production costs3. Factors affecting the cost of PHAs include raw materials availability, suitable process design, and downstream processing4,5. Since almost 50 % of the total production cos...
